蜕皮甾酮对大鼠脑内c-fos表达的影响

摘要 目的 观察大鼠海马内给予Aβ2 5 3 5后c fos基因在不同脑区表达的变化以及蜕皮甾酮的影响。方法 用立体定位仪向大鼠双侧海马内微注射Aβ2 5 3 5,免疫组织化学染色观察c fos基因的蛋白表达情况 ,采用BI 2 0 0 0图象处理系统中的免疫组化模块进行c fos表达的定量分析。结果 对照组的c fos在海马、丘脑及大脑皮层均有较高的表达 ;海马内注射Aβ2 5 3 5后 ,各脑区c fos基因的表达均明显降低 (P <0 .0 1) ;而蜕皮甾酮治疗组c fos基因的表达相对增加 :低剂量组 ,海马P <0 .0 5 ,丘脑P <0 .0 5 ;高剂量组 ,海马P <0 .0 1,丘脑P <0 .0 1,皮层P <0 .0 1。结论 海马内注射Aβ2 5 3 5后可明显降低海马、丘脑及大脑皮层c fos基因的表达 ;蜕皮甾酮可抑制Aβ2 5 3 5的作用 ,使海马、丘脑及大脑皮层的c Objective To observe the expression of c fos gene from brain of the rats induced by Aβ 25 35 and the effects of Ecdysterone on them. Methods Microinjection of Aβ 25 35 into the both hippocampus of the rat; The expression of c fos gene from the brain was detected by immunohistochemistry of ABC. Results There was a high expression of c fos gene in hippocampus, thalamus and cortex of the rat brain in GroupⅠwhile there was a much lowerexpression in Group Ⅱ(P<0.01); but in the treated ones of Group Ⅲ and Group Ⅳ, there was a relatively higher expression of c fos gene(P<0.05, P<0.01 respectively). Conclusions Aβ 25 35 microin jected into the hippocampus, lowered the expression of c fos gene from the brain of the rats in hippocampus, thalamus and cerebral cortices; Ecdysterone could inhibit it and therefore ECR relatively increased the expression of c fos gene from the brain of the rats in hippocampus, thalamus and cerebral cortices.
