摘要
目的 探讨大鼠脊髓损伤后白细胞介素 1 0 (IL 1 0 )、肿瘤坏死因子 α(TNF α)mRNA表达的时序变化规律。方法 SD大鼠 4 2只 ,随机分为 7组 ,采用改良Allen’s脊髓损伤打击模型 ,以逆转录 聚合酶链反应 (RT PCR)法测定伤段脊髓组织中IL 1 0、TNF αmRNA的表达情况。结果 正常脊髓组织内存在IL 1 0、TNF αmRNA的表达 ;脊髓损伤后IL 1 0mRNA表达逐渐上调 ,伤后 72小时明显上调 ,1 6 8小时达到高峰 ;TNF αmRNA伤后 1小时表达明显上调并达峰值 ,高表达持续至损伤后 2 4小时。结论 脊髓继发性损伤过程中抗炎性细胞因子与前炎性细胞因子共同存在 ;
Objective To study the changes gene of time course expression interleukin 10(IL 10) and tumor necrosis factor α(TNF α)gene after spinal cord injury(SIC) in rats. Methods Forty two SD rats were divided randomly into 7 groups. Modified Allen's model of spinal cord injury was established. Expression of IL 10 and TNF α mRNA after SCI were measured by reverse transcription polymerase chain reaction(RT PCR). Results Expression of IL 10 and TNF α mRNA were detected in non injured spinal cord. The expression of IL 10 mRNA was slowly up regulated after SCI,significantly up regulated at 72h,peaked at 168h post injury. The expression of TNF α mRNA significantly increased and peaked at 1h after SCI,and remained significantly higher than in the non injured spinal cord for 24h post injury. Conclusion There is coexistence of both antiinflammatory and pro inflammatory cytokines in the secondary SCI,and the antiinflammtory therapy of the secondary SCI should be used at early stage.
