广西眼镜蛇蛇毒细胞毒素的抗肿瘤作用 被引量:4

摘要 目的 了解广西眼镜蛇毒细胞毒素 (CTX )的抗肿瘤作用。方法 采用MTT法检测CTX对人鼻咽癌细胞株(CNE)、淋巴瘤细胞株 (YAC )、S180 癌细胞株、人宫颈癌细胞株 (HELA )和卵巢癌细胞株 (Ho8990 )体外细胞毒作用 ,同时应用动物移植性肿瘤的体内试验法 ,观察CTX对S180 载瘤小鼠 (S180 )和CNE的抑瘤作用及其生命延长率。结果 CTX对上述体外培养的肿瘤细胞有明显的细胞毒效应 ,作用 2 4h的半数抑制浓度分别为 1.0 4,0 .5 0 ,1.82 ,1.76和 1.0 5 μg/ml ;CTX对小鼠肿瘤有明显的抑制作用 ,抑瘤率为 12 %~ 3 7% ,呈明显量效关系。结论 CTX对体外培养的癌细胞株有较强的抑制作用 。 Objective To study the antitumor effect of Guangxi cobra venom cytotoxin on malignant tumor.Methods The cytotoxicity effect of CTX on human nasopharyngeal cancer cell(CNE),lymphoma(YAC) cell,sarcoma180 cell(S 180),uterus cervical carcinoma cell(HELA) and human ovarian carcinoma cell(Ho8990) was examined by MTT assay in vitro.The inhibitory effect of CTX was observed on the mice with transplanted tumor of CNE and sarcoma180(S 180) in mice.Results CTX had significant cytotoxicity effect on the above cancer cell lines in vitro.The IC50(24 hours in cubation) was 1.04,0.5,1.82,1.76 and 1.05 μg/ml for CNE,YAC,S 180,HELA,and Ho8990 cell,respctively.CTX exhibited significant antitumor activity on mice bearing nasopharyngeal and sarcoma180.The rate of inhibitor was between 12 and 37 percent.Inhibitory rate of CTX showed a dose-effect relationship.Conclusion CTX from Guangxi Cobra Venom exhibited strong cytotoxicity effect on cultural tumor cells line in vitro and inhibitory effect on mice bearing tumor.
