摘要
目的 观察氨磷汀 (amifostine ,化学代号 :WR 2 72 1)和峰龄多糖 (fenglingpolysaccharide ,FLPS)对人外周血粒 -巨噬祖细胞 (CFU GM )及HL60白血病细胞的化疗保护作用。方法 应用Ficoll分离单个核细胞 (MNC) ;甲基纤维素半固体培养法测定CFU GM集落 ;XTT法测定HL60细胞存活率 ;WR 2 72 1及FLPS处理的MNC ,与VP 16作用后 ,测定CFU GM集落产率 ,研究WR 2 72 1和FLPS对CFU GM的保护作用。结果 经WR 2 72 1和FLPS处理的MNC与VP 163 7℃作用 14h后 ,CFU GM集落产率均明显高于VP 16对照组 ,CFU GM集落数在VP 16对照组、空白对照组、WR 2 72 1+VP 16组、FLPS +VP 16组和WR 2 72 1+FLPS +VP 16组分别为每 1× 10 5个MNC 3 0 .9± 2 2 .5、83 .2± 43 .8、64 .6± 41.2、5 5 .3± 3 3 .5和 78.3± 48.2 ,与VP 16对照组相比 ,P值均 <0 .0 1;WR 2 72 1处理后的HL60细胞对VP 16的敏感性增强 ,IC50 由 5 2 .5 μg/ml下降为 40 .5 μg/ml ;VP 16对FLPS处理前后的HL60细胞的作用无明显改变。结论 WR 2 72 1和FLPS对人外周血CFU GM具有明显化疗保护作用。WR 2 72 1能增强HL60细胞对VP 16的敏感性。
Objective To observe the chemoprotective effects of amifostine(WR-2721) and Fengling polysaccharide(FLPS) on human peripheral blood granulocyte-macrophage progenitor cells(CFU-GM) and HL60 human leukemia cells.Methods Mononuclear cells(MNC) were deparated by Ficoll(1.077 g/ml).CFU-GM was measured by methylcellulose senisolid colony culture.HL60 cell survival rate were analyzed by XTT assay kit.MNC of WR-2721 treatment or containing FLPS were cultured 14h at 37 ℃ with VP -16.The cytoprotective activity against VP -16 toxic effects of WR-2721 and FLPS on CFU-GM were studied.Results The number of CFU-GM was significantly increased in MNC of adding FLPS or WR-2721 treatment.The mean value of CFU-GM in groups of VP-16 control,negative control,WR-2721+VP -16,FLPS+VP-16 and WR-2721+FLPS+VP -16 were 30.9±22.5,83.2±43.8,64.6±41.2,55.3±33.5 and 78.3±48.2 per 1×10 5,respectively.Compared with VP -16 control group,singnificant differences were seen between each group,P<0.01.After treatment with WR-2721,HL60 cells chemotherapy sensitivity to VP -16 was enhanced,and the IC 50 descended 52.5 μg/ml to 40.5 μg/ml.HL60 cells were cultured 14 h at 37 ℃ with 20 μg/ml VP -16 and 50 μg/ml FLPS,the cell survival rate was 88.0%±2.3%,negetive control was 90.5%±2.9%,no singnificant difference was seen between two group,n=21,P>0.05.Conclusion WR-2721 and FLPS selectiveiy protect human peripheral blood CFU-GM from the cytotoxicity of VP -16 without decreasing its cytotoxic effect on HL60 cells,and WR-2721 treatment could enhance HL60 cells chemotherapy sensitivity to VP -16.
