食管癌MMP-2、MMP-9及其组织抑制剂TIMP-1、TIMP-2的表达及临床意义 被引量:6

Expressions of MMP-2,MMP-9 and its Tissue Inhibitor TIMP-1,TIMP-2 in Esophageal Carcinoma and its Clinical Significance

摘要 目的 探讨食管鳞状细胞癌组织中 ,基质金属蛋白酶 (MMP)和金属蛋白酶组织抑制因子 (TIMP)的表达 ,及其与肿瘤浸润转移的关系。方法 应用免疫组织化学法 ,检测 45例食管鳞状细胞癌组织中MMP 2、MMP 9、TIMP 1、TIMP 2的表达情况。结果 MMP 2、MMP 9、TIMP 1、TIMP 2在食管鳞状细胞癌中表达阳性率分别为 80 .0 %、17.8%、13 .3 %和 84.4% ;不同分化程度癌组织的MMP、TIMP阳性表达率无显著性差异。随着肿瘤浸润的加深 ,MMP 2表达阳性率明显升高 ,有显著性差异 ( χ2=4.3 75 ,P <0 .0 5 )。有淋巴结转移者MMP 2表达阳性率明显高于无淋巴结转移者 ,有非常显著性差异 ( χ2 =6.2 5 2 ,P <0 .0 1)。结论 MMP 2、MMP 9主要在癌细胞中表达 ,TIMP 1、TIMP 2主要在间质细胞中表达。MMP 2、MMP 9、TIMP 1、TIMP 2表达与肿瘤分化程度无关 ,MMP Objective To investigate the expressions of matrix metalloproteinase-2,-9(MMP-2,MMP-9) and tissue inhibitor of metalloproteinase-1,-2(TIMP-1,TIMP-2) in esophageal carcinoma tissue and its correlation with invasion and metastasis.Methods MMP-2,MMP-9,TIMP-1,TIMP-2 proteins were detected by immunohistochemistry technique in 45 cases of esophageal squamous cell carcinoma(SCC).Results The positive rates of MMP-2、MMP-9、TIMP-1、TIMP-2 protein expression were 80.0%、 17.8%、13.3% and 84.4%.MMP-2,MMP-9 protein were mainly expressed in cancer cell,and TIMP-1,TIMP-2 in mesenchyma cell.The expressions of MMP-2、MMP-9、TIMP-1、TIMP-2 protein were not significantly different in cancer with different differentiation degree.The positive rate of MMP-2 expression was significantly higher in deeply invasive tissue than in superficially invasive tissue(χ 2=4.375,P<0.05),and it was also markedly higher in cases with lymph nodes metastases than in cases without(χ 2= 6.252,P<0.01).Conclusion MMP-2,MMP-9 protein are mainly expressed in cancer cells,and TIMP-1,TIMP-2 are mainly expressed in mesenchyma cells.The expressions of MMP-2、MMP-9、TIMP-1、TIMP-2 are not correlated with the cancer cell differentiation.The expression of MMP-2 is related to invasion and lymph nodes metastases of the cancer.
