链脲菌素性糖尿病大鼠模型对抗原引起的变应性气道炎症的影响 被引量:4

Effects of streptozotocin diabetes on allergen-induced airway inflammation

摘要 目的 :探讨 1型糖尿病对变应性气道炎症的干预作用及其机制。方法 :将 4 0只雄性SD大鼠分为 5组 ,每组 8只。A组 :腹腔注射链脲菌素 (STZ)建成 1型糖尿病模型 +卵清蛋白 (OVA)雾化吸入激发 ;B组 :腹腔注射OVA致敏 +雾化吸入激发 ;C组 :腹腔注射STZ +OVA致敏 +雾化吸入激发 ;D组 :OVA致敏 +腹腔注射STZ +激发 ;E组为对照组。检测各组血清C肽水平、Ⅳ型白细胞介素 (IL 4 )和γ 干扰素 (IFN γ)水平 ,作支气管肺泡灌洗液 (BALF)的细胞计数和分类 ;收获右下叶肺组织 ,切片并进行HE染色 ,光学显微镜下观察各组气道炎症细胞浸润情况。结果 :经过注射STZ干预的C组和D组的气道炎症、BALF的细胞总数及分类细胞均较B组显著降低 (P <0 .0 5 ) ,同时血清IL 4水平也较B组下降 (P <0 .0 5 ) ,IFN γ水平则增高 (P <0 .0 5 )。结论 :STZ性糖尿病可以抑制抗原引起的气道炎症过程 ,并通过调节辅助性T淋巴细胞 1(Th 1)和Th 2相关细胞因子的水平而实现。 Objective: To investigate the effect of type-1 diabetes on allergen-induced airway inflammation and it's potential mechanism. Methods: Forty male, 6 to 8 weeks age,SD rats were divided into five groups.Group-A:rats were render diabetes by injection of streptozotocin, and were challenged with an aerosolized solution of ovalbumin (OVA). Group-B:rats were sensitized, and challenged with OVA.Group-C:rats were render diabetes by STZ,and then sensitized, and challenged with OVA.Group-D:the OVA sensitized rats were made diabetes by STZ before OVA challenged.Group-E:nondiabetes, nonsensitized,only challenged.Bronchoalveolar lavage (BAL) was performed in rats from each group 24 h after challenged.Cells in BAL were counted and classified.Blood glucose were estimated,total and differential leukocyte counts were carried out in blood samples and serum were used for detection of IFN-γ,IL-4 and C-peptide. Results: The airway inflammation in both group-C and -D were less severe than that in the group-B.Total cells and eosinophil of BAL from the group-C and -D decreased significantly when compared with the group-B respectively( P <0.05). IL-4 expression were lower, while IFN-γ were higher in serum of group-C and group-D than that of group-B respectively ( P <0.05). Conclusion: type-1 diabetes induced by injection of streptozotocin might block allergen-induced airway inflammation and modulate the differentiation of T-helper lymphocyte .
