摘要
目的 观察鱼精蛋白对刺参酸性黏多糖 (SJAMP)抗凝血活性的对抗作用及其对SJAMP致死作用、血小板聚集、脾脏增重、抗癌活性的影响。方法 本实验测定内容包括 :凝血时间 (毛细管法 )、血小板计数 (显微镜法 )、脾脏重量、动物死亡率及动物肿瘤生长抑制率 (小鼠S180实体瘤 )。结果小鼠肌肉注射SJAMP 10 0mg kg ,凝血时间 ( 8.90± 1.85 )min ,血小板数量 ( 4 3 .40± 10 .16)× 10 1 1 个 L ,脾脏重量( 12 5 .68± 2 7.98)mg 10g ,肿瘤生长抑制率 3 2 .5 6%。小鼠肌肉注射SJAMP 10 0mg kg同时再腹腔注射鱼精蛋白 10 0mg kg ,其凝血时间 ( 3 .5 5± 1.5 2 )min(P <0 .0 1) ,血小板数量 ( 5 6.0 0± 5 .3 2 )× 10 1 1 个 L(P <0 .0 1) ,脾脏重量 ( 110 .2 0± 14 .0 9)mg 10g ,肿瘤生长抑制率 3 0 .81%。小鼠肌肉注射SJAMP 65 0mg kg ,注射后 2 4h及 48h动物死亡率分别为 9 2 0和 11 2 0 ,小鼠肌肉注射SJAMP 65 0mg kg ,再腹腔注射鱼精蛋白160mg kg× 2 ,相应的动物死亡率为 3 2 0 ( P =0 .0 3 4)和 5 2 0 ( P =0 .0 41)。结论 鱼精蛋白对SJAMP抗凝血活性及血小板聚集有明确的对抗作用 ,对SJAMP的致死性有部分保护作用 。
Objective To observe the effect of protamine (P)on biological activities of stichopus japonicus acid mucopolysaccharide (SJAMP).Method The tests included coagulative time (capillary tube method),mortality,platelet (microscopic counting method),weight of spleen and inhibition rate of tumor growth (murine solid tumor S180).Results The mice were given SJAMP 100 mg/kg im,the coagulative time was (8.90±1.85) min,the number of platelet was (43.40±10.16)×10 5/mm 3,the weight of spleen was (125.68±27.98) mg/10 g and the inhibition rate of tumor growth was 32.56%.The mice were given SJAMP 100 mg/kg im,adding P 100 mg/kg ip,the coagulative time was (3.55±1.52) min (P<0.001),the number of platelet was (56.00±5.32 )×10 5/mm 3(P<0.01),the weight of spleen was (110.20±14.09) mg/10 g and the inhibition rate of tumor growth was 30.81%.The mice were given SJAMP 650 mg/kg im,the mortality were 9/20 and 11/20 (24 h and 48 h after the administration respectively).The mice were given SJAMP 650 mg/kg im adding P 160 mg/kg×2 ip,the mortality was 3/20 (P=0.034) and 5/20 (P=0.041,24 h and 48 h after the administration respectively).Conclusion P has antagonistical effect on the anticoagulative effect and platelet aggregation induced by SJAMP in mice,P has some protective effect on the lethality induced by SJAMP in mice.P has no influence on immunoenhancement and anticancer effect of SJAMP in mice.
