摘要
目的 :观察球囊损伤后基质金属蛋白酶 9( MMP9)及其组织特异性抑制因子1 ( TIMP1 )在血管原位表达的动态变化 ,探讨其在球囊成形术后再狭窄 ( RS)形成中的作用机制。方法 :5 0只兔子随机分成正常组、术后 3、7、2 1、90天组 ,正常组不做处理 ,原位杂交方法观察 MMP9及 TIMP1的 m RNA改变。结果 :正常对照组 MMP9m RNA未见阳性表达 ;3d时 ,可见整个中层广泛阳性表达 ;7d时 ,新生内膜阳性表达 ,中层未见阳性表达 ;球囊损伤后随时间延长表达强度及范围逐渐减小 ;90 d时 ,未见阳性表达。正常对照组血管内皮细胞可见 TIMP1基础表达 ;3d时 ,中层广泛阳性表达 ;7d时 ,新生内膜阳性表达 ,中层未见阳性表达 ;2 1 d时 ,新生内膜阳性表达 ,中层未见阳性表达 ;90 d时 ,仍有新生内膜阳性表达。结论 :血管球囊成形术后 MMP9及 TIMP1 m RNA异常表达可能与血管平滑肌细胞迁移。
Objective: To investigate the altered expression of MMP9 mRNA, TIMP1 mRNA and it's possible function in restenosis after balloon injury. Methods: The expression of MMP9 and TIMP1 was examined day 3, day 7, day 21, day 90 after balloon injury in the carotid artery of rabbit with the method of in situ hybridization. Results: In situ hybridization demonstrated that the mRNAs for both MMP9 and TIMP1 were upregulated significantly on the medial three days after balloon injury and then decreased gradually. Only on the neointima did the MMP9 and TIMP1 mRNAs express one and three weeks after injury. TIMP1 mRNA continually expressed without expression of MMP9 on the neointima three months after injury. Conclusion: The abnormal expression of MMP9 mRNA, TIMP1 mRNA may correlate with vascular smooth muscle cells migration, neointima formation and constrictive remodeling after balloon injury.
