摘要
目的 观察异丙酚对人脐静脉内皮细胞(HUVECs)上细胞间粘附分子-1(ICAM-1)表达及对中性粒细胞(PMN)在内皮细胞上粘附数的影响。方法利用人脐带体外培养获得人脐静脉内皮细胞,将其随机分为空白组、异丙酚组、内毒素组、异丙酚+内毒索组。用流式细胞仪检测人脐静脉内皮细胞上ICAM-1的表达量。利用PMN分离液从人全血中分离出PMN。在光镜下计数PMN在内皮细胞上的粘附数。结果 4 μg/ml异丙酚对人脐静脉内皮细胞上ICAM-1的表达和对PMN在内皮细胞上的粘附数均无显著影响(P>0.05);40μg/ml异丙酚对人脐静脉内皮细胞上ICAM-1的表达和对PMN在内皮细胞上的粘附数均有抑制作用(P<0.01)。结论40μg/ml异丙酚对PMN与内皮细胞之间的过度粘附有抑制作用,从而可能对机体有一定的保护作用。
Objective To investigate the effects of propofol on the expression of intercelluar adhesion molecule-1 ( ICAM-1 ) on human umbilical vein endothelial cells ( HUVECs ) and the adhesion of poiymorphounclear cells (PMNs) to HUVECs. Methods HUVECs obtained form umbilical cord were cultured and randomly divided into four groups: control group (C); propofol group(PF); cndotoxin group(ET) and ET + PF group. Human PMNs were isolated from venous blood freshly obtained from 10 healthy volunteers. Propofol and/or endotoxin were added to the cultured HUVECs and incubated for 24 h. Then PMNs 1 μ 106 were added and incubated for 30 min. Flow cytometry was employed to detect the expression of ICAM-1 on HUVFCs. The neutrophils adhering to HUVECs were counted under microscope.Results Propofol 4 μg · ml-1 and 40 μg·ml-1 had no significant effect on ICAM-1 expression on HUVECs and also the number of PMNs adhering to HUVECs (P >0.05). Endotoxin 1 μg·ml-1 significantly increased the ICAM-1 expression and the number of PMNs adhering to HUVECs ( P < 0.01). Propofol 4 μg·ml-1 had no significant effect but 40 μg·ml-1 significantly decreased the rise in ICAM-1 expression and the number of PMNs adhering to HUVECs induced by endotoxin. Conclusion Propofol 40 μg·ml-1 can decrease the adhesion between neutrophils and HUVECs and the excessive inflammatory response. Thus it may be protective to the injured human body.
