摘要
目的 探讨肝移植术后急性排斥反应患者干扰素γ(IFN γ) ,白细胞介素 4 (IL 4 )和转化生长因子β(TGF β)的变化与相关性。 方法 急性排斥组与非排斥组按常规方法分离外周血单个核细胞 ,并用佛波脂、离子酶素和莫能酶素进行刺激。然后 ,采用双色流式细胞术分析移植后受者白细胞分化抗原 (CD) 4 + T细胞中IFN γ+ 、IL 4 + 和TGF β+ 细胞的百分率 ,并观察CD4 /CD8比值的变化。结果 肝移植术后非排斥组外周血CD4 + T细胞中IFN γ+ 、IL 4 + 和TGF β+ 细胞因子在第 7天分别为 (7 8± 1 4 ) %、(1 5± 0 6 ) %和 (0 9± 0 5 ) % ,第 14天分别为 (6 8± 1 6 ) %、(1 8± 0 8) %和(1 0± 0 6 ) % ,第 2 8天分别为 (7 0± 2 0 ) %、(2 0± 1 1) %和 (1 1± 0 6 ) % ,各组间差异无显著意义(P >0 0 5 )。移植后急性排斥组外周血CD4 + T细胞中IFN γ+ ,IL 4 + 和TGF β+ 细胞因子分别为(9 2± 1 5 ) %、(2 1± 0 6 ) %和 (1 5± 0 5 ) % ,与非排斥组相比 ,急排组IFN γ高表达 (P <0 0 5 ) ,而各组间IL 4和TGF β水平差异无显著意义 (P >0 0 5 )。 结论 Th1细胞因子的高表达与肝移植后急性排斥反应密切相关。
Objective To investigate the change of IFN-γ, IL-4, TGF-β in acute rejection after liver transplantation.Method Peripheral blood mononuclear cells(PBMC) were isolated and stimulated by PMA/ Ionomycin/ Monensin. IFN-γ, IL-4 and TGF-β production by CD4 + T cells were determined by using fluorescence activated cell sorter (FACS) analysis.Result The levels of IFN-γ, IL-4 and TGF-β were (7.8±1.4)%,(1.5±0.6)% and (0.9±0.5)% in the non-AR groups on 7 th hday, (6.8±1.6)%,(1.8±0.8)% and (1.0±0.6)%in the non-AR groups on 14 th day, and (7.0±2.0)%, (2.0±1.1)% and (1.1±0.6)% in the non-AR groups on 28 th day,respectively there were no significantly defferent in the non-AR groups on 7 th day,14 th day,28 th day ( P >0.05). The levels of IFN-γ, IL-4 and TGF-β were (9.2±1.5)%, (2.1±0.6)% and (1.5±0.5)% in the AR groups . comparation the AR with the non-AR groups on 7 th ,14 th ,28 th day respectively shows that high IFN-γ production was associated with AR in the early stage after transplantation period( P <0.05). There was no evidence of significant changes in IL-4 and TGF-β levels between non -AR groups with AR groups( P >0.05). Conclusion Th1 cytokine high production was significantly associated with acute rejection in liver transplantation recipients.
