摘要
目的 探讨病毒感染与趋化因子表达的改变在扩张型心肌病发病中的作用。方法利用RT PCR方法检测 12例原位异体心脏移植的扩张型心肌病受体心肌组织以及 5例正常对照心肌组织中Evs RNA的存在以及趋化因子RANTES、MCP 1、FKN及其受体US2 8的表达水平。结果在病理学证实的 12例扩张型心肌病心肌组织中有 7例Evs RNA阳性。2 5 % (3 12 )心肌组织中MCP 1mRNA的表达明显增高 ,经DNA同源序列分析 ,与人JEMCP 1同源性为 97%。而正常对照心肌组织中病毒基因及趋化因子检测均阴性。结论 肠道病毒感染可能会改变趋化因子的表达水平 ,而趋化因子的异常表达在扩张型心肌病发生发展中可能起重要作用。
Objective To study the effect of viral infection and chemokines in patients with dilated cardiomyopathy by observing the expression of chemokines in myocardium. Methods Myocardium specimens derived from 12 patients with dilated cardiomyopathy (DCM) underwent orthotopic cardiac transplantation and 5 normal myocardium specimens were collected at autopsy from sudden death as controls. Evs-RNA and expression level of RANTES、MCP-1、FKN and it′s receptor US28、 mRNA in those myocardium tissues were detected by RT-PCR. Results In 12 clinical suspected DCM patients, all of them were identified by pathology, and the positive Evs-RNA was in 7 patients. The expression of MCP-1 mRNA in 3 of 12 transplantation myocardium were markedly increased. After sequencing the cDNA, we found that their sequences were 97% homogeneity with human JE MCP-1. Moreover, in all 3 specimens of transplantation myocardium, Evs-RNA were detected. Neither viral gene nor the expression of MCP-1 was found in myocardium of control group. Conclusion Viral infection can alter the expression level of chemokines MCP-1. The abnormal expression of chemokines may play an important role in DCM development.
