灵芝(孢子粉)生药质量检测 被引量:2

摘要 目的 通过显微形态、化学成分的检测分析 ,探讨灵芝孢子粉生药质量检测方法。方法 一般化学成分按常规方法进行 ,锌、锰等微量元素分析采用原子吸收分光度法 ( AAS) ,多醣含量的检测采用苯酚 -硫酸比色法 ,牛磺酸分析采用氨基酸自动分析仪 ;破壁形态观察和破壁率的检测采用数字显微镜、扫描电镜进行。结果  破壁前后孢子的粗蛋白、总醣、微量元素、牛磺酸等含量均变化不大 ,而粗脂肪 (索氏提取 )及多糖 (热水提取 )含量明显增加。在扫描电镜和光学显微镜下 ,未破壁孢子粉的显微形态完整 ,而破壁孢子粉样品中观察不到完整的孢子 ,其孢壁结构基本解体 ,孢内物质是易释出。结论   ( 1)水溶性多糖 ( Polysaccharides)、灵芝酸 ( Triterpenoid acids)及牛磺酸 ( Taurine)等含量可以作为灵芝孢子粉生药质量控制的重要指标 ;( 2 ) OBJECTIVE In this paper,micro morphology and chemical components were tested and analysed to research the quality examination and detection methods on the spore(Crude drug)of Ganoderma lucidum.METHODS the general chemical components micro elements,polysaccharide and taurine(Tau)were analysed by conventional methods,atomic absorb spectrophotometer(AAS),phenol vitriol colorimtety and amino acids auto analyzer respectively.Micro morphology of the intact and wall creaked spores were observed by scanning electron microscope(SEM)and and digital optical microscope(DOP).RESULTS the contents of protein total sugar and some micro element(Zn,Mn,Cr,Ni)and taurine between the intact and wall cracked spore,were relatively steady.But the contents of fats(Soxhlat extraction)and polysaccharide(water solubility)were increase d obviously.Under SEM and DOP,there wrew not completed morphology among the wall creaked spores that the ultrastructure of sporoderm were destroyed and the endo substance were easily to be dispersed or extract,while the intact spores were intactness(ovoid).CONCLUSION (1)Polysaccharide and taurine could be considered as the important target componets for quality control on the spore(Crude drug)of Ganoderma lucidum,(2)The morphology of wall creaked spores could be observed and examined and the sporederm broken rates also could be detected by scanning electron microscope(SEM)and digital optical microscope(DOP).
