摘要 【目的】探讨自发性高血压大鼠 (SHR)颈动脉血管间质胶原重构的特点及应用厄贝沙坦逆转的意义。【方法】选择 1 6周龄雄性SHR 2 4只分为SHR组、厄贝沙坦组 (30mg·kg-1 ·d-1 ) ,另选同龄雄性Wistar大鼠 1 2只为正常对照组 ;颈总动脉切片采用HE染色、苦味酸 -天狼星红 (PSR)染色分别检测颈动脉壁厚 /腔径比值 (W /C)、胶原面积百分比 (VFC)。【结果】SHR组W /C值较正常对照组高 (P <0 .0 1 ) ,厄贝沙坦组治疗 2 0周后 ,较SHR组下降 (P <0 .0 1 ) ;SHR组胶原面积百分比较正常对照组高 (P <0 .0 1 ) ,厄贝沙坦组治疗 2 0周后 ,较SHR组也有下降 (P <0 .0 5 ) ;SHR组血管壁厚 /腔径比值和胶原面积百分比均与SBP相关(均P <0 .0 5 ) ;厄贝沙坦组血管壁厚 /腔径比值与SBP相关 (P <0 .0 5 ) ,而胶原面积百分比则与血压的相关性不明显。【结论】SHR颈动脉血管存在以壁厚 /腔径比值和胶原面积百分比增高为特点的重构 ,应用厄贝沙坦时血管壁厚 /腔径比值能逆转 。 To investigate the characteristics of vascular collagen remodeling of the carotid artery and significance of irbesartan to reverse the effect in spontaneous hypertension rats (SHRs).Twenty-four male SHRs (age 16 wks) were selected and divided into 2 groups:①SHR group,12 SHRs; ②Irbesartan group, 12 SHRs were treated with irbesartan in a dosage of 30 mg·kg -1 ·d -1 . In addition, 12 male Wistar rats with the same age served as ③ Normotensive control group. The vascular volume fraction of collagen (VFC) and the ratio of carotid artery wall thickness /cavity diameter (W / C) were detected by picrosirius red and HE staining, respectively.In SHR group, VFC and W / C were significantly higher than those of controls (both P<0.01); these two indexes were correlated to their systolic blood pressure(SBP)(P<0.05).Twenty wks after treatment, VFC and W/C in irbesartan group decreased obviously (P<0.01) and considerably(P<0.05) as compared with those in SHR group; but correlation of the former to blood pressure was not so prominent, while the latter was correlated with SBP(P<0.05).[Conclusion]A remodeling with characteristics of increased VFC and W/C exists in the carotid artery of SHR. Irbesartan is beneficial to reverse the W/C ratio of carotid artery in SHR , but it can only reverse their VFC to certain extent.
