摘要
目的 :研究成人骨髓间充质干细胞在体外培养条件下 ,传代次数与细胞凋亡的关系。方法 :采用An nexinV/PI双染色法 ,在流式细胞仪上检测不同传代数的成人骨髓间充质干细胞的凋亡情况。结果 :传代次数越多 ,骨髓间充质干细胞的凋亡率及死亡率越高 ,存活的正常细胞越少。结论 :第 4代以内的细胞增殖能力强 ,凋亡率低 。
Objective:To investigate the relationship between apoptosis rate of the adult MSCs cultured in vitro with the times of subculture passage Method:The apoptosis rate of adult MSCs within different passages was investigated by flowcytometry with Annexin V/PI Result:The more times MSCs was subcultured,the rate of apoptosis and death was higher and the fewer alived Conclusion:The MSCs within 4 passages should be used as the seeding cells of tissue engineering because of strong proliferative ability and low rate of apoptosis
