摘要
研究了用两步法由芴合成氯甲酸 9 芴甲酯 (Fmoc Cl) ,即第 1步由工业芴用氢化钠还原法制备 9 芴甲醇 ;第 2步由 9 芴甲醇与双光气合成Fmoc Cl。当工业芴 83.0g与氢化钠 6 5 .0 g ,甲酸乙酯 5 0 0mL反应时 ,芴甲醇收率达 87.0 % ;当双光气与 9 芴甲醇摩尔比为 6∶1,反应时间 8h ,反应温度 5 0~ 6 0℃时 ,Fmoc Cl收率达 90 .2 %。
The synthesis of 9-fluorenyl methoxy carbonyl chloride(Fmoc-Cl) from flourene by 2 steps was studied, i.e. the 1st step is to synthesize the 9-fluorenyl methanol from industrial fluorene and the 2nd step is to synthesize Fmoc-Cl from 9-fluorenyl methanol. When flourene 83.0 g, NaH 65 g and ethyl formate 500 mL were used in the 1st step, the yield of fluorenyl methanol was 87.0%; and when the molar ratio of diphosgene to 9-fluorenyl methanol was 6∶1, reaction time was 8 h, reaction temprature was 5060 ℃ in the 2nd setp, the yield of Fmoc-Cl was 90.2%.
