摘要
目的 :检测兔颈髓急性牵张性损伤后细胞凋亡及相关蛋白Bax和Bc1 2的表达 ,探讨颈髓急性牵张性损伤神经细胞凋亡的特点。方法 :兔颈髓经不同程度轴向牵张损伤后 ,分别在 2、 4、 8、 2 4和 72h处死取材。应用末端脱氧核苷酸转移酶介导生物素标记技术 (Tunel法 )检测细胞凋亡 ,应用免疫组化技术对脊髓组织进行标记 ,以检测Bc1 2和Bax的表达。结果 :损伤 4h后Bax大量表达 ,在 2 4h达到高峰 ,与凋亡细胞的出现基本平行 ,而Bc1 2仅有少量表达。结论 :兔颈髓急性牵张性损伤后细胞凋亡可能是早期主要表现之一。凋亡相关蛋白Bax大量表达 ,并可能在神经细胞的凋亡过程中起重要作用。
Objective:To study the character of neuron apoptosis after acute stretch injury in rabbit cervical spine through detecting apoptosis and the expression of its correlative proteins (Bax and Bc1-2). Method:Forty-two Japanese rabbits were randomly divided into 4 groups and subjected to a controlled different degree axial traction to cause an acute C 5~6 spine injury,then rabbits were killed at 2,4,8,24 or 72 hours after injury(n=6).The corresponding segments of cervical spine were cut for morphological studies including terminal desoxynucleotidyl transferase-mediated dUPT nick end labeling(TUNEL) and immunohistochemical staining et al. Result:Bax expresssd at 4h after cervical spinal injury,it reached its peak at 24h while Bc1-2 expressed low. Conclusion:Neuron apoptosis may be one of major manifestations early after acute stretch injury in rabbit cervical spine. There is high expression of apoptosis correlative protein(Bax),it may play an important role in induction of neurons to apoptosis.
