9个STR位点荧光法检测与血型血清学方法鉴定亲子关系的对比研究 被引量:1

摘要 目的 对血型血清学与PCR -STR荧光检测这两种鉴定亲子关系的实验方法进行对比研究。方法 常规亲子鉴定中 ,采用血型血清学方法检查ABO、Rh、MN、P、HLA -A、B等血型 ,PCR -STR荧光法分析D8S1 1 79等 9个STR位点长度多态性。结果 ①在 1 66例亲子鉴定中 ,认定亲生关系的为 1 32例 ,排除亲生关系的 34例 ;② 1 32例认定亲生关系案例中 ,血清学方法RCP值介于 83 .40 %~ 99.9% ,平均 96 .2 7% ;而PCR -STR方法RCP值介于 99.82 96 %~ 99.99998% ,平均 99.986 %。③ 34例排除亲生关系的案例中 ,血清学方法排除 33例 ,排除的血型系统和HLA位点为 1~ 4个 ,平均 1 .8个 ;而PCR -STR方法排除 34例 ,排除的STR位点为 3~ 9个 ,平均 5 .8个 ;血清学不排除而STR位点排除亲生关系的案例有 1例。④观察 2 64次减数分裂 ,发现FGA、A7S82 0位点各突变 1次。结论 本文的 9个STR位点的非父排除概率高于血清学方法。当 9个STR位点均符合遗传规律时 ,RCP值通常可达 99.99% ;但STR位点存在基因突变 ,需增加其它的遗传标记进行检测 ,以保证结果的准确。 Objective To make a comparative study of serological method and PCR-STR method used in paternity testing. Methods In routine patemity testing, the blood groups of ABO,Rh,MN,P and HLA-A,B locus were detected by serological method the fragment size of nine STR loci were detected and genotyped by automatic DNA sequencer.Results ①Among 166 cases of paternity testing were detected, 132 cases were proved with demonstrated and 34 cases were demonstrated with no-paternity. ②In the 132 cases with demonstrated patemity, the RCP value for serological method and PCR-STR genotyping method were at the range of 83.4%~99.99% and 99.8296%~99.99998% respectively. ③We observed 34 no-paternity cases, an average number of 1.8 exclusions at HLA-A B locus and other blood group were found in 33 cases,but the average number of STR cxclusions were 5.8 in all no-paternity cases. One case that can not be excluded from paternity by serological method was demonstrated with no-paternity by PCR-STR genotyping method. ④We observed 264 meiosis, single STR mutation was observed in two cases. Conclusions The accumulated PE value for 9 STR loci is much high than serological method. When the genotypes of 9 STR loci were in accordance with Mendel inheritance,the RCP value of most paternity can reach 99.99%or more, When one or two STR mutations were observed, other more genetic marders must be detected before making a final conclusion.
