摘要
籽粒硬度是重要的小麦(T.aestivum L.)品质性状。水洗淀粉表面分子量约15 KDa的Friabilin蛋白是这一性状的生化基础。用单粒谷物特性仪(SKCS)和改进的聚丙烯酰胺凝胶电泳(SDS-PAGE)分析了104份国内外冬小麦品种(系)的籽粒硬度与Friabilin表达量的关系,结果表明,Friabilin谱带相对表达量与籽粒硬度显著相关,Friabiiin谱带强,籽粒硬度数值低,Friabilin谱带弱,籽粒硬度数值高;用Friabilin蛋白表达量判断籽粒软硬的准确率为86.5%,其相关系数为-0.66,达到1%的显著水平。因此,改进的Friabilin蛋白SDS-PAGE技术可用于小麦籽粒硬度生化标记的辅助选择。
Grain hardness is important quality trait of common wheat (T.aestivm L) . A starch surface associated protein, friabilin (Mr15KDa), is a putative marker for softness. 104 varieties and advanced lines were used to measure kernel hardness and friabilin level by Single Kernel Characterization System (SKCS) and improved SDS-PAGE. The results show that high level of friabilin protein in soft wheat and relatively low level in hard wheat, with correlation coeffi-cience-0.66 for the tested materials. An improved SDS-PAGE method as a biochemical marker assistant selection for grain hardness in common wheat.
