摘要
以文心兰杂交新品种星战、达卡、爪哇的茎尖及花蕾为组培材料,对适合外植体的原球茎增殖、分化的培养基、培养方式进行试验研究。对培养基激素进行筛选试验结果表明:利于原球茎诱导增殖的培养基为1/2MS+6-BA5 0rng/L+NAA0 5mg/L;利于不定芽增殖的培养基为1/2MS+6-BA2 0mg/L+NAA0 1mg/L;利于生根的培养基为1/2MS+NAA0 5mg/L。其试管苗移植的栽培基质以小石子+小树皮(1 1)为佳。并从控制温、湿度,采取合理的水肥管理措施,进行病害防治等方面,介绍了文心兰组培苗的移植管理技术。
Stem apex and flower buds of new variety Xingzhan, Daka and Zhuawa, hybridization from Oncidium hybridum, as material of tissue culture, the experiment on medium favorable for explant propagation and differentiation and culture methods was conducted. The result of hormone selection of medium indicates that the medium favorable for explant propagation inducing is 1/2 MS + 6-BA 5.0mg/L + NAA 0.5mg/L, that favorable for adventitious bud propagation is 1/2MS + 6-BA2.0mg/L + NAA0.1mg/L and that favorable for rooting is 1/2MS + NAA0.5mg/L. The optimum cultivation stroma for transplant of test-tube plantlets is pebbles adding some small pieces of barks. The paper also introduces transplant and management techniques of Oncidium hybridum tissue culture plantlets from aspects of control temperature and humidity, applying water and fertilizer rationally and disease and insects control.
