摘要
目的 研究山苦茶提取物对豚鼠离体胆囊肌条的作用及作用机制。方法 将肌条置于恒温浴槽,以BL-410生物功能实验系统记录肌条张力变化。结果 山苦茶提取物对离体胆囊肌条具有剂量依赖的收缩作用,最大效应为乙酰胆碱(10μmol·L^(-1))的76%±8%。EC_(50)为0.14~0.30 mg·mL^(-1)(n=8),阻滞药吲哚美辛和维拉帕米显著降低其缩胆囊作用。结论 山苦茶提取物可通过增加前列腺素的释放以及直接作用于钙通道对豚鼠离体胆囊产生收缩作用。
OBJECTIVE To investigate the effects and the mechanisms of mallotus furetianus on the gallbladder smooth muscle in vitro in guinea pigs. METHODS The gallbladder strips were mounted in organ baths containing 20 ml. Krebs solution at 37℃, and the isometric tension was measured using a force transducer (JH-2) and recorded with Experimental System of Biological Function (TME, China). RESULTS Mallotus furetianus had concentration-dependent contractile effect on gallbladder strips of the guinea pigs. The maximal effect was 76%±8%(n=8) com pared with acetylcholine and the EC_(50) value was 0.15~0.26 mg·mL^(-1), which could be significantly reduced by cyclooxygenase inhibitor indomethacin and calcium blocker verapamil. CONCLUSIONS Mallotus furetianus is a potent contractile stimulant in guinea pigs, whose effect is related to cyclooxygenase and calcium channal.
