摘要
目的观察红景天苷对乙醛刺激的肝星状细胞(HSC)凋亡的诱导作用,以及此过程中c-Jun氨基末端激酶(c-JunN-terminalkinase,JNK)活性的变化。方法体外培养大鼠HSC株;用不同浓度的红景天苷(1.0、1.5、2.0mg/mL)对乙醛(200μmol/L)刺激的大鼠HSC进行处理,采用流式细胞仪检测细胞凋亡率;Westernblot法检测磷酸化JNK(p-JNK)蛋白水平。结果不同浓度的红景天苷对乙醛刺激的HSC中p-JNK的水平有明显下调作用(平均光密度比值分别为35.8±3.4、24.9±2.7、3.4±0.9),强度呈剂量依赖关系,同乙醛组(MVI值为48.6±4.8)相比有显著性差异(P<0.05);同时具有促进细胞凋亡的作用:空白对照组及乙醛组中细胞凋亡率为2.0±0.5%、11±2.0%,两者有极显著性差异(P<0.01),经红景天苷处理后细胞凋亡率均上升(分别为27±3.2%;33±2.5%;53±3.5%),与乙醛组相比有显著性差异(P<0.05)。结论红景天苷可以诱导乙醛刺激的HSC凋亡,下调JNK活性可能是其诱导HSC凋亡的机制之一。
Objective To investigate the effect of Salidroside in inducing apoptosis of rat hepatic stellate cells (HSC) stimulated by acetaldehyde and to observe the changes of c- Jnk N- terminal kinase (JNK) activity.Methods HSC stimulated by acetaldehyde were cultured in vitro and were treated with different concentrations of Salidroside.Apoptotic rate was analyzed by flow cytometry and the activity of phosphorylating JNK was measured by Western blot method.Results Salidroside in different concentrations (1.0,1.5,2.0 mg/mL) suppressed the activity of JNK in a dose- effect manner.Average light density was 35.8± 3.4,24.9± 2.7 and 3.4± 0.9 in Salidroside groups, which differed from that in acetaldehyde group( 48.6± 4.8; P < 0.05).It could also increased apoptotic rate of HSC (P < 0.05 as compared with acetaldehyde group).Conclusion Down- regulating JNK binding activity may be one of the cellular mechanisms of Salidroside in inducing the apoptosis of HSC stimulated by acetaldehyde.
