喜树茎段不定芽的诱导及再生系统的建立 被引量:7

摘要 选用1年生喜树带芽茎段为外植体,筛选出了喜树最佳诱导培养基为MS+NAA0.05mg/L+6-BA1.0mg/L,诱导率为92%;芽增殖的最佳培养基是MS+NAA0.1~0.5mg/L+BA1.0mg/L,分化频率为82%~87%;壮苗培养基为MS+NAA0.05mg/L+BA0.3mg/L;最佳生根培养基为1/2MS+NAA0.1mg/L+IBA1.0mg/L,生根率为95%。初步建立了喜树无性微繁再生系统,并对喜树外植体选择及最佳生根条件进行了讨论。 Adventitious buds were induced using the first year shoot with a bud of Camptotheca acuminata as explant and cultured on MS and B5 medium supplemented with various concentrations of hormones NAA and BA. In our test range, the result showed that MS+NAA 0.05 mg/L+6-BA 1.0 mg/L is the best medium for inducing the development of axillary buds, with 92% success; MS+NAA 0.1~0.5 mg/L+BA 1.0 mg/L is the best for bud multiplication, with differentiation frequency of 82%~87%; MS+NAA 0.05 mg/L+BA 0.3 mg/L is optimal for strengthening the plantlets, whereas 1/2MS+NAA 1.0 mg/L+IBA 1.0 mg/L is the best medium for rooting induction, the rooting rate is up to 95%.
