双向电泳分析二烯丙基二硫诱导HL60细胞分化的蛋白表达差异 被引量:5

摘要 目的 以二烯丙基二硫 (DADS)诱导人急性早幼粒细胞性白血病HL60细胞分化为模型 ,利用双向凝胶电泳 ( 2 -DE)技术分析比较DADS诱导HL60细胞分化的蛋白表达差异。方法 细胞形态学观察与NBT还原反应结合验证HL60细胞的分化 ;用双向电泳技术分析蛋白表达差异。结果 分化后细胞形态发生明显变化 ,细胞核变小 ;NBT还原能力明显增强。双向电泳技术分析筛选了 32个差异点。结论 双向电泳技术可直接反映细胞蛋白表达的变化 。 s: Objective To analyze the protein expression in myeloid leukaemia cell line HL60 with diallyldisulfide treatment. Methods Cell differentiation was validated by morphologic observation and nitroblue tetrazolium (NBT) reduction test. The differences of protein expressions in HL60 cells treated with 8.5×10 -6 mol/L diallyldisulfide was analyzed through the results from two-dimensional electrophoresis. Results HL60 cells were induced to differentiate into granulocytes with diallyldisulfide. The morphology of the treated cells was significantly different from control, and NBT reduction in treated cells was increased. By two-dimensional electrophpresis, 32 protein spots were detected to be different. Conclusion The change of proteins expression in cells was directly refected by two-dimensional electrophoresis. The expressions of some proteins are increased and some reduced in HL60 cells after diallydisulfide treatment.
