摘要 目的 :应用碱性单细胞凝胶电泳 (SCGE)对脑梗死患者急性期、恢复期血液中单个核细胞的DNA损伤与修复情况进行检测 ,判断脑梗死患者DNA损伤水平与病程的关系 .方法 :急性脑梗死患者 2 4例 ,分别在发病 5d内及 2 5~ 2 8d抽取空腹静脉血 3mL ,1 .5mL用于SCGE检测 ,在荧光显微镜下观察单个核细胞的彗星率并分析慧尾DNA的含量 .1 .5mL分离血清 ,- 2 0℃保存 ,用于神经元特异性烯醇化酶 (NSE)测定 ,同时选取年龄、性别匹配的健康者 2 0例作正常对照 .结果 :脑梗死患者急性期血液中大多数单个核细胞出现彗星样改变 ,恢复期患者血液中彗星率下降 ,而同龄健康对照者血液中大多数单个核细胞表现为圆形 ,仅少数表现为略带拖尾的荧光改变 .急性期脑梗死患者NSE含量较对照组显著增加 (P<0 .0 1 ) ,而恢复期脑梗死患者NSE含量与对照组无显著差别 .结论 :自由基及DNA损伤修复在脑梗死的发生。 AIM: To investigate the DNA damage of mononuclear cells in the peripheral blood of cerebral infarction patients at acute and recovery stages and to evaluate the correlation between the DNA damage and the pathologic course of cerebral infarction. METHODS: Twenty four patients with cerebral infarction and 20 age and sex matched healthy persons were selected as cases and controls, respectively. The damaged DNA was assayed by single cell gel electrophoresis (SCGE) or comet assay, and the neurone specific enolase (NSE) was measured by radioimmunological methods. RESULTS: The rate of comet cells and their tail DNA concentration increased significantly in acute cerebral infarction patients, and so did the NSE concentration. The percentage of comet cells and tail DNA decreased significantly in convalescence patients, but no difference in the NSE concentration was found between recovered patients and control group. CONCLUSION: The free radical and DNA damage and repair may play an important role in the pathogenesis, development and recovery of cerebral infarction.
