摘要
目的 研究细胞增殖核抗原 (PCNA)在雌激素诱导的SD大鼠垂体泌乳素腺瘤中的表达及其意义。方法 2 4只雄性SD大鼠随机分为雌激素组及对照组 ,雌激素组大鼠局麻后 ,分别于大鼠背部植入长 1cm ,直径 1 5mm的硅胶管 (药泵 ) ,管中预装 10mg的 17 β雌二醇 ,而对照组用同样的方法植入同样大小未装药的硅胶管。随后饲养 4个月 ,断头取出垂体 ,称重 ,同时采血进行血清泌乳素 (PRL)测定 ,垂体石蜡切片进行PCNA检测。结果 雌激素组大鼠重量皆大于 30mg ,明显高于对照组 (P <0 0 1) ,PRL明显高于对照组 (P <0 0 1) ,PCNA表达阳性。而对照组垂体皆小于10mg ,PCNA阴性表达。结论 PCNA是可取代PRL检测来判定垂体泌乳素腺瘤动物模型建立的一个指标。
Objective To study the espression and significance of PCNA in prolactinomas in Sprague-Dawley(SD)rat.Methods Firstly 24 SD rats were randomly divided into control groups and estrogenic groups.The rats of estrogenic groups were embeded a silic tube in the skin which was 1 cm in length and 1.5 mm in diameter.The tube was packed with 10 mg of 17-β estradiol.The control groups was embedded in the same way an empty silic tube.Then those rats were breeded for 3 months.Then the pituitary was weighed and PRL and PCNA was assayed.Results The weight of rat in estrogenic groups was 30 mg or more and higher than that in control groups( P< 0.01)and PRL was higher than in the control groups( P< 0.01)and PCNA positive expression was found.In contrast the ratsweight in the control groups was less than 10 mg and PCNA negative expression was found. Conclusion PCNA could replace the assay of PRL in the SD rat animal models as an important parameter which was induced by estradiol.
