恶性黑色素瘤血管生成拟态的体内外实验和超微结构研究 被引量:4

摘要 目的:通过动物实验、细胞三维培养及电镜观察恶性黑色素瘤组织中血管生成拟态的形成过程及其超微结构特点。方法:建立恶性黑色素瘤B16移植瘤动物模型和恶性黑色素瘤细胞株LiBr的三维培养模型,研究恶性黑色素瘤组织血管生成拟态,同时用透射电镜观察人体恶性黑色素瘤组织血管生成拟态的超微结构特点。结果:动物实验证实恶性黑色素移植瘤模型内存在血管生成拟态;细胞三维培养模型结果表明恶性黑色素瘤细胞存在肿瘤细胞自身变形并与胶原蛋白粘附、肿瘤细胞彼此围成通道结构的现象,电镜观察证实了血管生成拟态的存在及其结构。结论:恶性黑色素瘤组织内存在血管生成拟态,肿瘤细胞可以通过血管生成拟态结构获得充足的血液供应。 Objective: Vasculogenic mimicry was studied in malignant melanoma by constructing tumor cell tri-dimension culture system, animal experiment and trans-electron microscope. Methods: To construct tumor cell tri-dimension culture system and animal experiment malignant melanoma B16 model, observe the morphologic diversification structure and blood supplyment of malignant melanoma where vasculogenic mimicry was investigated. Those tumors which exist vasculogenic mimicry was observed by electron microscope. Results: Animal experiment and electron microscope proved that vasculogenic mimicry exists in malignant melanoma. Tri-dimension culture model of malignant melanoma cell line also testified the tumor cells self-metamorphose and vasculorized trend. Conclusion: Vasculogenic mimicry exists in malignant melanoma. The tumor cell can obtain oxygen and nutriment through this structure.
