摘要
目的 研究人早孕期绒毛及滋养细胞趋化因子SDF 1的表达、分泌及其对蜕膜免疫活性细胞的募集作用。方法 收集早孕期绒毛组织并检测SDF 1的定位表达 ;用ELISA分析早孕期滋养细胞培养上清液中SDF 1的浓度水平 ;分离早孕期蜕膜免疫活性细胞 ,以趋化试验研究SDF 1及滋养细胞培养上清液对蜕膜免疫活性细胞的趋化作用。结果 人早孕期滋养细胞表达并分泌SDF 1;一定浓度范围的SDF 1对蜕膜免疫活性细胞具有趋化作用 ,最大趋化指数为 3 .2 7± 0 .89;滋养细胞培养上清液亦明显趋化蜕膜免疫活性细胞 ,趋化指数为 2 .11± 0 .79。结论 人早孕期滋养细胞表达的SDF 1对蜕膜免疫活性细胞具有趋化、募集作用 ,从而有助于形成蜕膜独特的淋巴细胞群体并维持母
Objective To investigate the expression and the secretion of chemokine SDF-1 in human first trimester trophoblasts and to exami ne the chemotactic activity of SDF-1 of decidual lymphocytes. Method s We detected the expression of SDF-1 by in situ hybridization and immunohistochemistry. We also assessed the presence of SDF-1 in the culture of isolated trophoblasts derived from human first trimester villi. The chemotaxis o f SDF-1 to decidual lymphocytes was tested in Transwell and the chemotactic act ivity was quantitatively examined. Results Both in situ h ybridization and immunohistochemistry experiments showed that human first trimes ter trophoblasts expressed SDF-1. Highly purified trophoblasts were found by EL ISA to secrete SDF-1 in vitro. SDF-1 exhibited chemotactic activity of dec idual lymphocyte, and the maximum CI(chemotactic index) was 3.27±0.89. Supernat ants in the trophoblast cell culture were also found to be chemoattractant to de cidual lympocytes with CI as 2.11± 0.79 . Conclusion H uman first trimester trophoblasts produced SDF-1, which in turn endowed trophob lasts with the capacity to attract decidual lymphocytes. This activity contribut ed to the recruitment of decidual lymphocytes and may manipulate the microimmune environment at the materno-fetal interface.
