摘要
目的 观察 D-半乳糖老化小鼠海马神经元 β-淀粉样肽 (Aβ)及其相关蛋白表达水平的变化以及 APP1 7肽对其改变的影响。方法 采用免疫组织化学染色和免疫印迹方法 ,检测 D-半乳糖老化小鼠模型海马神经元 Aβ、β-分泌酶、内质网 Aβ结合蛋白和早老蛋白 - 1的表达水平 ,并观察应用 APP1 7肽的保护作用。结果 对照组小鼠海马神经元 Aβ及其相关蛋白表达量少 ,而 D-半乳糖老化小鼠海马神经元 Aβ及其相关蛋白表达水平明显增加 ,与对照组比较差异有高度显著性 (P<0 .0 1 ) ,APP1 7肽组上述各种蛋白的表达结果与对照组接近。结论 D-半乳糖老化小鼠海马神经元存在着 Aβ及其相关蛋白表达的上调 ,APP1 7肽可使之恢复正常水平 。
Objective To investigate the changes of β-amyloid (Aβ) and its related protein of D-galactose induced aging of murine brain (DGAM) and the effect of APP17-mer peptide on the variation Methods The expression level of Aβ, beta-site APP-cleaving enzyme (BACE), endoplasmic reticulum amyloid β-peptide binding protein (ERAB) and presenilin-1 (PS-1) of DGAM were studied with immunohistochemical staining and western blotting, and the protective effect of APP17-mer peptide was observed Results The expression level of Aβ and its related protein in the hippocampal of DGAM increased obviously compared with control group (P<0 01), while the result of APP17-mer peptide group was the same as those in the controls Conclusions The expression of Aβ and its related protein in the hippocampal of DGAM are up-regulated, while APP17-mer peptide normalizes the expression and protects neurons
