小檗碱对人胃癌细胞增殖、细胞周期及CD44V6表达的影响 被引量:31

摘要 目的 :研究小檗碱对人胃癌MGC 80 3细胞增殖、细胞周期及CD4 4V6分子表达的影响 ,探讨其抗肿瘤及抗肿瘤转移作用机制。方法 :MTT法检测小檗碱对人胃癌MGC 80 3细胞体外增殖作用。激光共聚焦显微镜观察用药后细胞形态的改变并用流式细胞仪检测细胞周期的变化。应用免疫组化和流式细胞术技术检测药物对细胞表面CD4 4V6表达的影响。结果 :小檗碱对MGC 80 3细胞有显著抑制作用 ,在 10、2 0、4 0 μg ml浓度下 ,其抑制率分别为 36 2 %、4 9 7%和 5 9 3% ,有明显剂量依赖关系。激光共聚焦显微镜下细胞核固缩 ,并可见凋亡小体。药物可将细胞阻滞在G0 G1期 ,并使细胞表面的CD4 4V6表达降低。结论 :小檗碱可通过诱导MGC 80 3细胞凋亡并将细胞阻滞在G0 G1期 ,发挥抑制瘤细胞的体外增殖作用 ;小檗碱可降低MGC 80 3细胞株CD4 4V6的表达 。 Objective:To study the effected of berberine on cell proliferation,cell cycle and CD44V6 expression in gastric cancer cell(MGC-803).To investigate the mechanism of inhibitory tumor proliferation and tumor metastasis.Methods:MTT assay was used to determine the cell proliferation effected by berberine.The change of cell morphology and cell cycle were detected by laser scanning confocal microscope(LSCM) and flow cytometry,respectively.Immunohistochemical staining and flow cytometry were applied to detect the CD44V6 expression on cell surface.Results:It was found that the proliferation of MCG-803 is inhibited by berberine.The inhibitory rate of drug(10、20、40 μg/ml)is 36 2%、49 7% and 59 3% respectively in a dose-dependent manner.The cell nuclear condensation and formation of apoptotic bodies were found under LSCM.The cell cycle were arrested in G0-G1.The expression of CD44V6 on cell surface was obviously decreased.Conclusion:Berberine could inhibit proliferation of MGC-03 by inducing apoptosis and arresting cell in G0-G1.Berberine could inhibit tumor metastasis by decreasing the expression of CD44V6 on MCG-803.
