龈下菌斑涂片两种染色分类方法的比较

Comparison of two staining methods for the same subgingival plaque samples

目的 :用革兰染色和刚果红负染两种不同的染色方法对同一龈下菌斑样本进行分类计数 ,比较两种分类方法的优缺点 ,提出应用中的问题以及解决方法。方法 :随机抽样采集 4 0例牙周病患者的 80个位点龈下菌斑 ,同一标本进行生理盐水涂片革兰染色和刚果红负染。光学显微镜油镜 (15× 10 0 )镜检共计数 2 0 0个细菌 ,包括 5种不同形状细菌。采用SPSS 10 0 1统计软件 ,对数据进行分析。结果 :两种染色分类方法计数统计结果为螺旋体、弯曲菌、梭形菌 3种菌数值P >0 0 5无统计学意义 ,而球菌、杆菌 2种菌数值经统计P <0 0 1有高度统计学意义。结论 :刚果红负染简便易行 ,但龈下菌斑球菌、杆菌进行百分计数时 ,不适宜用刚果红负染法 ,应根据实验目的选择恰当的方法。

Objective:To compare difference of two bacteria staining methods and problems during its use. Methods:Subgingival plaque samples of 80 sites (2 site/person)from 40 cases of periodonititis were collected.The plaque samples were smeared on glass slides with 0.9% saline solution and stained by Gram staining.The 2% Congo red solution was used as another staining method.A total of 200 bacteria of five species were counted including spirochaeta,coccus,bacillus,fusiformis and campylobacter. Results:There were no statistical differences among spirochaeta,fusiformis and campylobacter between methods.The others were different significantly.Conclusion:The Congo red negative staining method is easy to use and to keep the bacteria were readily to be identified.However the Congo red negative staining method may be interferd by granules from plaque during smear.
