摘要
目的 观察银杏叶提取物一金纳多(Extract ofGinkgo Biloba Leaves Injection,EGB)对抗顺铂(Cisplatin,CDDP)耳毒性的作用。方法 将24只豚鼠随机分为CDDP组、EGB+CDDP组及对照组,采用听性脑干反应(auditory brainstem response,ABR)、血清超氧化物歧化酶(superoxide dismutase,SOD)活性和丙二醛(MDA)含量、光镜及扫描电镜技术,观察用药前后各指标的变化。结果CDDP组动物ABR阈值较其它两组明显升高(P<0.01),其余两组间差异亦有显著性(0.01<P<0.05)。血清SOD活性和MDA含量检测表明,CDDP组血清SOD活性较对照组明显降低(P<0.01),EGB+CDDP组较对照组则差异不显著(P>0.05)。血清MDA含量以CDDP组升高最显著(P<0.01),EGB+CDDP组较对照组略有升高,没有显著性差异(P>0.05)。耳蜗扫描电镜显示EGV+CDDP组较CDDP组毛细胞受损程度明显减轻。结论EGB可有效减轻CDDP的耳毒性作用。
OBJECTIVE To investigate the protective effects of Extract of Ginkgo Biloba Leaves Injection(EGB) against ototoxicity of Cisplatm(CDDP). METHODS 24 guinea pigs were randomly devided into three groups:CDDP, EGB+CDDP and control.Auditory brain stem response(ABR), light microscopy, scanning electron microscopy, activity of SOD and content of the metabolite of lipid peroxidation(MDA) in serum were tested to evaluate the effect on hearing threshold, serum level and morphology. RESULTS the hearing threshold (ABR) was significantly higher in CDDP group than that in EGB +CDDP group (P<0.01). The vitality of SOD in serum was significantly lower in CDDP group than in the control group(P<0.01).However the difference between the other two groups was not signicant(P>0.05). The content of the metabolite of lipid peroxidation(MDA) in serum was significantly(P<0.01) higher in CDDP group than that in other groups.The structural damage of hair cells was more severe in CDDP group than in other groups.CONCLU-SION Extract of Ginkgo Bioba leaves injection(EGB) can effectly reduce ototoxicity of cisplatin.
