盐酸特拉唑嗪诱导人前列腺间质细胞凋亡的机制研究 被引量:8

摘要 目的 探讨盐酸特拉唑嗪诱导人前列腺间质细胞凋亡的机制。方法 在原代体外培养的人增生前列腺间质细胞中加入不同浓度的盐酸特拉唑嗪、转化生长因子 (TGF) β1及TGF β1抗体 ,用原位凋亡法 (TUNEL)检测细胞的凋亡状况 ,TGF β1酶联免疫粘附试验 (ELISA)试剂盒检测细胞悬液A值。结果 盐酸特拉唑嗪和TGF β1均可诱导体外培养的人前列腺间质细胞凋亡 ,且具有浓度依赖性 ;加入特异性TGF β1抗体后细胞凋亡率减少 72 .7% (P <0 .0 1) ;盐酸特拉唑嗪组细胞悬液TGF β1浓度的A值较对照组高 87.5 % (P <0 .0 1)。 结论 盐酸特拉唑嗪诱导人前列腺间质细胞凋亡通过或至少部分通过细胞自分泌TGF β1引起。 Objective To explore the mechanism of apoptosis of human prostatic stromal cells induced by terazosin-HCl.Methods The primary culture of benign prostatic hyperplastic cells was carried out for 3 generations.Different concentrations of terazosin-HCl,TGF-β1 and TGF-β1 specific antibody were added to the cell medium.The apoptosis was assessed by TUNEL assay.The TGF-β1 concentration was determined by enzymelinked immunosorbent assay (ELISA).Results The apoptosis of human prostatic stromal cells could be induced by terazosin-HCl in a concentration-dependent manner (r=0.896, P<0.01).The apoptosis in the group of terazosin-HCl plus TGF-β1 specific antibody was increased by 72.7% as compared with that in the terazosin-HCl group (P<0.01).The apoptosis of human prostatic stromal cells could also be induced by TGF-β1 and positively correlated with the concentration of TGF-β1 (r=0.732,P<0.01).The A value,which represented the concentration of TGF-β1,in the terazosin-HCl group was decreased by 87.5% as compared with that in the control group (P<0.01).Conclusion The mechanism of apoptosis of human prostatic stromal cells induced by terazosin-HCl is mediated through,or at least partially,an autocrine production of TGF-β1.
