不同速率生长的胶质瘤细胞的葡萄糖代谢状态研究 被引量:2

Relationship between growth rate of glioma cell and glucose uptake

摘要 目的 观察不同速率生长的胶质瘤细胞的葡萄糖代谢状态。方法 用C6细胞克隆培养得到不同细胞形态、不同生长速率的细胞株。分别检测生长最快的C64细胞和最慢的C61细胞的胞内葡萄糖、ATP浓度 ,同时检测两种细胞的己糖激酶蛋白和它们的胞质 /线粒体己糖激酶活性。结果 C64细胞内检测不到葡萄糖 ,C61细胞内葡萄糖浓度为 (0 .17± 0 .0 5 ) μmol/g蛋白 ;C64细胞、C61细胞内ATP浓度分别为 (10 .16± 1.5 1) μmol/g蛋白和 (7.5 7± 0 .3 2 ) μmol/g蛋白。条带的吸光度分析显示C64细胞的己糖激酶Ⅰ稍高于C61细胞 ;C64细胞线粒体己糖激酶活性约为C61细胞的 3倍 ,但其胞质己糖激酶活性仅为C61细胞的一半。结论 葡萄糖转运系统与肿瘤细胞的葡萄糖摄取、葡萄糖代谢状态及生长速度等密切相关。 Objective To investigate glucose metabolic status of C6 glioma cells at different growth rates.Methods C6 glioma cells were subcloned to produce different cell lines differing in morphology,growth rate:C64 cells grew rapidly whereas C61 cells grew slowly.The intracellular glucose concentration and ATP concentration of both cell lines were measured respectively,and the levels of hexokinase protein,cytosolic as well as mitochondrial hexokinase activities were determined.Results The intracellar glucose concentration of C64 cells was undetectable,whereas that of C61 cells was (0.17±0.05) nmol/mg.protein.The intracellar ATP concentrations of C64 cells and C61 cells were (10.16±1.51) nmol/mg.protein and (7.57±0.32) μmol/g.protein respectively.Densitometric analysis of the bands showed that the total amount of hexokinase protein in C64 cells was only slightly higher than that in C61 cells.The mitochondrial hexokinase activity of C64 cells was about three times higher than that of C61 cells,but the cytosolic hexokinase activity of C64 cells was about half that of C61 cells.Conclusion The glucose transport system is closely related to the glucose uptake,the glucose metabolic status and the growth rate of glioma cells.
