强直性脊柱炎患者外周血单个核细胞干扰素诱导蛋白10及Th1/Th2型细胞因子的表达

The expression of interferon induced protein 10(IP-10) and Th1/Th2 cytokines in the peripheral blood mononuclear cells of ankylosing spondylitis patients

目的 探讨干扰素诱导蛋白 10 (IP 10 )在强直性脊柱炎 (AS)发病中的作用。方法 收集肝素抗凝的AS患者的外周全血 ,梯度离心得到外周血单个核细胞(PBMC) ,应用半定量RT PCR法测定IP 10、IFN γ和IL 4mRNA表达水平。结果 AS患者PBMC中IP 10、IFN γ和IL 4mRNA表达水平均较健康对照组高(P <0 0 1) ,同时IFN γ/IL 4值也较对照组高 (P <0 0 5 )。IFN γ和IL 4mRNA表达水平与C 反应蛋白呈负相关 ,IP 10mRNA表达水平与IFN γ、IL 4mRNA和IFN γ/IL 4值均呈正相关。结论 AS患者PBMC表达的细胞因子以Th1型为主 ,IP 10表达升高 ,IP

Objective To investigate the role of IP-10 in the pathogenesis of ankylosing spondylitis (AS). Methods The peripheral blood monocytic cells (PBMC) were collected from 39 AS patients, and the mRNA expression of IP-10, IFN-γ and IL-4 were measured by semi-quantitive RT-PCR with G3PDH as the internal control. Results The mRNA expression levels of IP-10, IFN-γ and IL-4 as well as the rate of IFN-γ/IL-4 were elevated in the PBMC of AS patients compared with the healthy controls (P<0.05). The levels of IFN-γ and IL-4 levels were negatively correlated with C reactive protein, while the level of IP-10 was positively correlated with that of IFN-γ, IL-4, and the rate of IFN-γ/IL-4. Conclusions The mRNA expression of Th1 cytokines was dominant in the PBMC of AS patients, and IP-10 mRNA level was elevated obviously. The chemokine IP-10 might take part in the pathogenesis of AS through facilitating homing of Th1 cells to the inflammatory site.
