摘要
目的 :验证所构建的组织型纤溶酶原激活剂 (tPA)乳腺特异性表达载体pWTA能否在小鼠乳腺组织中瞬时表达。方法 :采用直接注射法将pWTADNA、pWTADNA/脂质转染胺试剂LipofectamineTM2 0 0 0 复合物及PBS分别注射入妊娠末期小鼠乳腺组织。应用原位杂交技术 ,以地高辛标记的tPAcDNA为探针 ,观察pWTA在小鼠乳腺组织中的瞬时表达。结果 :tPA表达阳性细胞散在分布于pWTADNA及pWTADNA/LipofectamineTM2 0 0 0 复合物注射小鼠部分乳腺腺泡 ,PBS注射组无阳性细胞。结论
Aim: To demonstrate the expression of pWTA in mammary gland tissues of mice. Methods: The pWTA DNA,pWTA DNA/ Lipofectamine TM2000 complex,and PBS were directly injected into mammary gland tissues of pregnant mice,respectively. The tPA transient expression in mouse mammary gland was demonstrated by in situ hybridization using Dig labeled tPA cDNA probe. Results: The positive cells were scattered in some acini of mouse mammary gland of experimental groups, and no positive signals were exhibited in control group. Conclusion: The pWTA can specificly express tPA in mouse mammary gland.
