摘要
目的 :从新生大鼠海马齿状回分离并鉴定神经干细胞。方法 :利用无血清方法分离、培养新生大鼠齿状回神经细胞 ,采用免疫细胞化学方法检测神经巢蛋白 (Nestin)并鉴定神经元和神经胶质细胞。结果 :从新生大鼠海马齿状回分离的细胞群可不断增殖形成神经球 ,并能自我更新 ,表达Nestin。自神经球分化出的细胞分别表达神经元、星形胶质细胞特异性抗原。结论
Aim: To isolate and identify neural stem cells (NSCs) from dentate gyrus of hippocampus of neonatal rats. Methods:Serum free medium and sequence proliferation of neurospheres were utilized to isolate,incubate neural cells from dentate gyrus of neonatal rats. Immunocytochemistry was applied to detect Nestin antigen.Results: The cell group isolated from dentate gyrus of hippocampus of neonatal rats possessed the ability of proliferation and self renew.Nestin positive cells could be found in the cell groups. Cells after differentiation could express certain antigen of astrocytes and neurons.Conclusion:The neural stem cells from the fascia dentate gyrus of hippocampus have the ability to undergo self renew and multiproliferation.
