高三尖杉酯碱诱导HL60细胞端粒酶活性的变化

摘要 目的 :研究高三尖杉酯碱 (Homoharringtonine ,HHT)对HL6 0细胞端粒酶活性的影响。方法 :不同浓度HHT作用于HL6 0细胞 1 2h及 0 .0 2mg/LHHT作用HL6 0细胞不同时间后 ,采用TRAP PCR ELISA检测HL6 0细胞端粒酶活性。结果 :HHT浓度增加至 0 .0 4mg/L和 0 .0 5mg/L时 ,HL6 0细胞端粒酶活性与 0mg/L组相比明显下降 (P均 <0 .0 5 )。 0 .0 2mg/LHHT作用HL6 0细胞 1 8h、2 4h、30h后 ,HL6 0细胞端粒酶活性明显低于 0h组 (P均 <0 .0 5 ) ,且低于空白对照 (P均 <0 .0 5 )。结论 :HHT是一种有效的端粒酶活性抑制剂 。 Aim: To investigate telomerase activity in HL60 cells exposed to homoharringtonine(HHT). Methods: HL60 cells were exposed to HHT at different concentrations for 12 hours,or exposed to 0.02 mg/L HHT for different time,then telomerase activity in HL60 cells was detected by TRAP PCR ELISA. Results: Compared with the control (0 mg/L HHT), telomerase activity in HL60 cells reduced significantly when exposed to 0.04 mg/L and 0.05 mg/L HHT ( P < 0.05 )and compared with the control (0 h),telomerase activity reduced significantly when exposed to 0.02 mg/L HHT for 18 h,24 h and 30 h ( P <0.05). Conclusion: HHT is a kind of telomerase inhibitor. Down regulation of HHT on induced telomerase activity in HL60 cells is in a time and dose dependent manner.
