摘要
目的 :研究白细胞介素 (IL) 1β在大鼠心脏移植排斥反应中心脏局部的表达变化情况 ,探讨其在心脏移植急性排斥反应诊断中的可能作用。方法 :建立大鼠颈部异位心脏移植模型 ,不同品系大鼠之间的移植供、受体分别为SD大鼠和Wistar大鼠 (共 5 0只 ) ,同品系大鼠之间的移植供受体均为Wistar大鼠 (共 5 0只 )。观察移植心脏自然生存时间 ,另于术后第 1、3、5、7、9、11天 ,分别切取移植心脏各 5只 ,提取总RNA ,进行逆转录 ,将目的引物和对照引物在同一管中扩增 ,以两者像素值比值均数作曲线图 ,观察排斥反应时IL 1βmRNA的动态表达情况。结果 :不同品系移植心脏的停跳高峰出现在术后第 6~ 10天。排斥反应时移植心脏局部IL 1βmRNA的表达水平明显增高 ,峰值在术后第 1天。结论 :IL 1β在心脏移植排斥反应中可能起重要的作用 ,IL
Objective:To detect the expression and changes of IL-1β in the rats heart transplantation, and assess the effect IL-1β on the acute rejection in rats heart transplantation. Method:A simplified heterotopic heart transplantation model, inbred SD and wistar rats were used as donors and recipients respectively in allogeneic transplantation. In synogeneic transplantation, both the donors and recipients were Wistar rats. Twenty allografts and twenty isografts were assessed daily by palpation to observe graft's viability. In every time point, five allografts and five isografts were used to detect IL-1β mRNA expression. The allografts and isografts were harvested on postoperative days 1, 3, 5, 7, 9, 11. After the total RNA was prepared, the control primer and target primer were amplified in the same tube. The average ratio of the target products' pixel volume to control's was calculated, by which the coordinates chart was made to detect the expression level of IL-1β mRNA directly. Result:The levels of IL-1β mRNA expression in allografts were much higher than those in isografts. The peak of elevated level of the IL-1β mRNA expressions appeared on the first day post-transplantation, and prior to the peak of the allografts dead time. Conclusions:The experiment suggested that IL-1β may play an important role in rejection of heart transplantation. The determination of IL-1β mRNA expression may serve as a useful immunologic method for diagnosing acute rejection.
