摘要
目的 :观察反义c-myc寡核苷酸对大鼠胸腺淋巴细胞增殖的抑制作用。方法 :Ficoll密度梯度离心法分离大鼠胸腺淋巴细胞。利用Iipofectin将正义、反义及错配c -myc寡核苷酸导入大鼠胸腺淋巴细胞 ,[3 H]-TdR掺入法及MTS法检测淋巴细胞增殖 ,RT -PCR检测c-mycmRNA的表达。结果 :反义c-myc寡核苷酸可抑制大鼠胸腺淋巴细胞的增殖 [(0 14± 0 0 3)Avs (0 32± 0 16 )A ,P <0 0 5 ],但此抑制作用无浓度依赖性 ,反义c -myc寡核苷酸可降低胸腺淋巴细胞c-mycmRNA的表达。结论 :反义c -myc寡核苷酸可抑制大鼠胸腺淋巴细胞的增殖。
AIM: To observe the antiproliferative effect of c-myc antisense oligonucleotide in rat thymus lymphocytes. METHODS: Rat thymus lymphocytes were separated by Ficoll-Urografin density gradient centrifugation. Lipofectin was used to introduce antisense, sense and mismatched oligonucleotides for c-myc to rat thymus lymphocytes. The antiproliferative effect was assayed by incorporation of -TdR and MTS cell proliferation assay. TR-PCR was used to detect the expression of c-myc mRNA. RESULTS: c-myc antisense oligonucleotide inhibited rat thymus lymphocytes proliferation [(0.14±0.03)A vs (0.32±0.16)A, P<0.05], but this effect had no relationship with the concentration of c-myc antisense oligonucleotide. c-myc antisense oligonucleotide decreased the expression of c-myc mRNA in rat thymus lymphocytes. CONCLUSION: c-myc antisense oligonucleotide inhibited rat thymus lymphocyte proliferation.
