摘要
目的 确证槐角染料木素的化学结构并研究其药理学作用。方法 ①以核磁共振、红外光谱、质谱和紫外光谱对槐角染料木素的结构进行鉴定。②♀SD大鼠随机分为 6组 :对照组 (假手术 +生理盐水 ) ;模型组 (去卵巢 +生理盐水 ) ;尼尔雌醇组 (去卵巢 +尼尔雌醇 ) ;大剂量组 (去卵巢 +染料木素 18mg·kg-1) ;中剂量组 (去卵巢 +染料木素 9mg·kg-1) ;小剂量组 (去卵巢 +染料木素 4 5mg·kg-1)。在手术后d 8开始灌胃给药 ,每日 1次 (尼尔雌醇组每周 1次 ) ;分别于术后第 0、1、2、4、8、12周从各组各取 10只大鼠 ,处死取骨作骨密度、骨Ca2 + 、P3 + 、Mg2 + 和组织形态学测定。结果 ①四谱技术测定数据确证 ,槐角染料木素的化学结构为5 ,7,4′ 三羟基异黄酮。②动物实验结果表明 ,染料木素在中、小剂量可明显增强模型大鼠股骨和胫骨的骨密度 ,特别在第 4周与模型组比较差异有显著性 (P <0 0 1) ;小剂量组可明显升高骨Ca2 + 水平 ,其它各用药组均可升高骨P3 + 、Mg2 + 水平 ;中、小剂量组可明显增加模型大鼠的骨小梁面积百分比 (%Tb .Ar)和骨小梁厚度 (Tb .Th)。结论 槐角染料木素与市场销售的染料木素化学结构相同 ;槐角染料木素在中、小剂量组具有预防去势大鼠骨质丢失的作用 。
AIM The aim was to study the chemical structure and the pharmacological action of genisitein from Huaijiao. METHODS (1) Chemical structure purified genisitein was examined by using the nuclear magnetic resonant spectrum, infrared absorption spectrum, ultraviolet absorption spectrum and mass spectrum.(2) SD female rats were divided randomly into 6 groups,including control,model, nilestriol,highdosage(18 mg·kg -1),medial dosage(9 mg·kg -1), and low dosage(4.5 mg·kg -1) of genisitein.The animals in the control were sham-operated and given saline solution. The rats ovaiotomized of other five groups were given saline, nilestriol and differential dosage of Huaijiao genistein separately.The rats were fed with different drugs by intragast once a day,once a week for the nilestriol group,on the eighth day after surgery.The animals (10 rats) were sacrificed in 0,1,2,4,8,and 12 weeks after surgery, and the rats tissues were processed for analysis of bone density,levels of bone calcium, phosphorus, magnesium and histomorphology. RESULTS (1)The chemical structure of Huaijiao genistein is 5,7,4′-trihydroxyisoflavone.(2) Compared with the controls, the animals treated with Huaijiao genistein at a dosage of 4.5 mg·kg -1 or 9 mg·kg -1 had (P<0.01) higher density of femur and tibia than the model group, especially by the fourth week after treatment. The low dosage genistein group had higher bone calcium, and the other treatment groups had higher bone phosphorous and magenesium (P<0.05). In addition, the groups of low or medial dosage genistein were obserred of increased trabecular thickness( Tb.Th) and trabecular area percentage(%Tb.Ar)(P<0.01). CONCLUSION Chemical structure of the Huaijiao genistien is the same as that of genistein in the market. Treatment with a low or medial dosage of Huaijiao genistein will prevent osteoporosis, but the effects decrease at high dosage (18 mg·kg -1).
