摘要
目的 观察正常人成纤维细胞老化过程中β 半乳糖苷酶的变化规律。 方法 取正常人二倍体成纤维细胞进行传代培养 ,以不同代次的细胞为实验对象 ,通过细胞生长曲线、群体倍增时间和 β 半乳糖苷酶染色等一系列检测方法 ,对成纤维细胞老化过程进行了观察。结果 随着成纤维细胞的连续传代培养 ,SA β Gal表达呈现从弱 (在年轻细胞中占 4 % )到强 (在老化细胞中占 6 0 % )的趋势。
Objective To investigate the alteration of the level of β-galactosidase in the replicative senescence of normal human dipoid fibroblasts(HDF).Methods Specimens were taken for cell culture.The aging process was displayed by growth curve of cell line and β-galactosidase histochemical staining.Results The expression of SA-β-Gal tended to be more apparent in old cells than that in young cells,which maintained relatively as low as about 4%.Conclusion The alteration of SA-β-Gal contributes to creating cellular senescence aging pattern in vitro.
