羚羊角和羚羊角塞针剂中氨基酸和多肽成分的分析

Analysis of Amino Acid and Polypeptide in Cornu Saigae Tataricae and Cornu Tampon Saigae Tataricae Injection

利用柱前衍生HPLC法 ,以异硫氰酸苯酯 (PITC)为衍生试剂分离并测定羚羊角和羚羊角塞针剂中氨基酸含量。实验结果表明羚羊角及其角塞针剂中都富含脯氨酸、甘氨酸、丝氨酸等 1 6种氨基酸。用HPLC从两种针剂中分离出近 2 0个色谱峰 ,对其中 6个较大的峰用基质辅助激光解吸与离子化时间飞行质谱法测得两种针剂中均存在 4组分子量差别较大的小分子多肽成分 ,分子量分别为 1 30 0± 2 0 ,1 0 70± 2 0和 830± 30 ,6 80± 2 0Da。从另一个角度验证了羚羊角及其角塞针剂中含有相似的成分 ,具有解热、抗惊和镇静作用 。

The contents of amino acid in Cornu Saigae Ataricae and Cornu Tampon Saigae Tataricae Injection were detected by HPLC with PITC as reagent. The result showed that there are abundant 16 kinds of amino acids such as proline, aminoacetic acid, serine in Cornu Saigae Ataricae and Cornu Tampon Saigae Tataricae Injection. There are 20 pieces of chroma peak by HPLC from the two kinds of injection, in which 6 pieces of bigger chroma peaks were detected by mass spectrometry with base assisted laser ionization. It showed that there are 4 groups little molecule polypeptide with large difference in weight in both Injection. The molecule weights were 1300±20,1070±20, 830±30 and 680±20Da differently. It verified that Cornu Saigae Ataricae and Cornu Tampon Saigae Tataricae Injection has similar components in another way. It has the effect of relieving heat and sedation, which offers experiment basement to the determination and development of the effective components of Cornu Saigae Ataricae and Cornu Tampon Saigae Tataricae Injection.
