摘要
以香菇( Lentinus edodes )菌丝为材料,以纤维素酶、β-葡萄糖醛酸苷酶的组合酶溶液降解菌丝细胞壁,获原生质体。香菇菌丝原生质体在含1.5g/L酵母膏、20g/L葡萄糖与0.6mol/L甘露醇的液体培养基中培养6—8h后,开始启动生长。培养24h,可观察到分枝状菌丝的形成。定点连续观察表明,不论是单向出芽生长还是双向出芽生长的再生方式,都是原生质体在培养中正常的生长方式。单向生长的芽管状菌丝、双向生长的菌丝以及多分枝的菌丝形态,是原生质体再生过程中不同生长阶段的表现。
Protoplasts were harvested from the mycelia of Lentinus edodes by digesting the wall with the mixed enzyme solution made up of cellulase and β-glucuronidase.After cultured them in the medium composed of 1.5 g/L yeast extract, 20g/L glucose and 0.6 mol/L manitol for 6-8 h, the protoplasts started germinating.Formation of the branchy mycelia was observed within 24h in culture.Successive observation for one protoplast in situ showed that both the germination types of the protoplasts, whether unilateral or branchy,are normal.They were the different morphological expression during the procedure of the protoplast growth.
