不同预处理对小麦幼胚抗除草剂基因转化效率的影响 被引量:1

Effect of different pre-treatment on the frequency of transformation of herbicide-resistant gene of wheat embryo

摘要 以小麦品种豫麦18号和豫麦34号作为供试材料,用基因枪介导法进行了抗除草剂基因的转化,研究了不同预培养时间和高渗处理对小麦幼胚转化效果的影响.结果表明,幼胚以诱导培养10~15d的转化效果最好;用0 4mol·L-1的甘露醇在轰击前6h和轰击后18h对10~15d预培养的幼胚进行处理,可明显提高转化效率. Two wheat cultivars,Yumai 18 and Yumai 34 were used as experiment materials to study the effect of different pre-culture duration of immature embryo and osmotic treatment on tranformation frequency by transforming herbicide-resistant gene through particle baombardment.The result showed that the immature embryos pre-cultured for 10~15 days and treated with 0.4 mol·L^(-1) mannitol could significantly improve gene transformation frequency.
