实验性消化道溃疡小鼠小肠上皮内淋巴细胞及其亚群IL-2及IFN-γ分泌活性的变化

摘要 目的 :了解消化道溃疡小肠上皮内淋巴细胞 (iIEL)及其亚群 (Thy1 -)IL 2和IFN γ的分泌活性。方法 :利用水浸限制刺激建立小鼠应激性消化道溃疡模型 ,用ELISA方法分别检测总iIEL和Thy1 -iIEL分泌的IL 2和IFN γ的含量。结果 :给予水浸限制刺激后 1d总iIEL产生的IL 2水平与对照组比较显著性上升 (P <0 .0 5 ) ,6d下降到正常水平 ;IFN γ于 3d升到最高 ,与对照组比较升高显著 (P <0 .0 1 ) ,4d开始下降 ;Thy1 -iIEL所分泌的IL 2和IFN γ的水平虽然表现为轻度升高 ,但与对照组比较无显著性差异 (P >0 .0 5 )。结论 :iIEL可能通过分泌IL 2、IFN γ等细胞因子参与消化道溃疡的修复过程 ;IL 2和IFN γ可能主要是由Thy1 + Objective: To understand changes of secreting activities of IL 2 and IFN γ in intraepithelial lymphocytes(iIEL) and its subset(Thy1 - iIEL) in alimentary tract ulcer. Methods: ELISA was used to examine secreting levels of IL 2 and IFN γ in total iIEL and Thy1 - iIEL in the animal model with digestive ulcer induced by water immersion restrain stress. Results: The IL 2 level produced by total iIEL was elevated significantly on the 1st day after the alimentary tract ulcer was induced (P<0.05) and was declined to normal on the 6th day. The IFN γ level was elevated to the peak on the 3rd day and started to be decreased on the 4th day, which was also statistically significant compared with that of the control groups(P<0.01). Both IL 2 and IFN γ levels produced by Thy1 - iIEL showed no significant change compared with those of the control groups(P>0.05).Conclusion: The iIEL may play a role in repairing the alimentary tract ulcer by secreting cytokines such as IL 2 and IFN γ. The IL 2 and IFN γ may be mainly produced by Thy1 + iIEL.
