应用酶组织细胞化学技术对真空冷冻干燥保存兔角膜内皮细胞的研究 被引量:2

摘要 目的 :探讨真空冷冻干燥保存法对于兔角膜内皮细胞活性的影响。方法 :将冷冻及冻干保存兔角膜片分别复水及复温后 ,与新鲜角膜同时行内皮细胞三磷酸腺苷酶 (ATPase)、琥珀酸脱氢酶 (SDH)的酶组织细胞化学染色 ,观察 3组酶活性的差异。结果 :3组样本ATPase及SDH酶组化染色均呈阳性反应。实验组与对照组间AT Pase和SDH酶的平均积分光密度值 (IOD)差异具有统计学意义 (P <0 .0 1 )。结论 :真空冷冻干燥保存法可使离体角膜组织保持一定的活性。与新鲜及冷冻角膜相比 。 Objective: To investigate the influence of vaccum freeze drying preservation(VFDP) on the activity of cornea endothelial cells of rabbits. Methods:Before usage the VFDP cornea was rehydrated and the cryopreservated cornea was thawed. We detected the activity of Na +,K + ATPase and succinate dehydrogenase(SDH) by using enzyme histochemistry in corneal endothelial cells of rabbit. Results:The enzyme histocytochemistry reaction of Na +,K + ATPase and SDH of all groups were positive. The differences of Na +,K + ATPase and SDH activity were significant (P<0.01). Conclusion:The VFDP can keep the corneal endothelium available. Compared with fresh cornea, the activity of VFDP cornea is lower. But the VFDP might become a kind of new method to preserve cornea for long time.
