银杏细胞固定化培养及银杏内酯的产生 被引量:6

摘要 探讨利用银杏 (GinkgobilobaL .)细胞固定化培养方法 ,大规模生产药用成分银杏内酯类化合物的可能性。考察了植物生长调节剂对银杏固定化培养细胞生长及发酵液中银杏内酯类成分产生的影响。结果显示 ,最优培养基生长调节剂配比为 2 ,4 D 8 0mg L +KT 0 0 4mg L +NAA 0 4mg L。优化培养后 ,固定化细胞最高生物量出现在第 1 2d ,为 0 4gDW 瓶。发酵液中银杏内酯含量 :GKA在 2 2d时最高 (2 0 0 μg L) ,GKB在 1 8d时最高 (1 2 2 μg L) ,GKC在 1 8d时最高 (5 0 9μg L)。 Cultures of immobilized cells of Ginkgo biloba L,as well as the production of their secondary metabolite--Ginkgolides were reported in this paper.The effects of plant growth regulators on the growth of immobilized cells cultures and the production of ginkgolides were determined.The result showed that the optimized combination of growth regulators is 2,4-D 8.0?mg/L+KT 0.04?mg/L+NAA 0.4?mg/L.The highest biomass (0.4?g DW/bottle) of immobilized cultured cells appeared in twelfth day,the highest content(200?μg/L) of GKA in culture solution in twenty-second day,GKB(122?μg/L) and GKC(509?μg/L) in eighteenth day.The content of ginkgolides of optimized hormone treatment in the immobilized culture medium was much higher than that of control culture.
