浙南汉族人群短串联重复序列LPL位点的多态性调查

摘要 目的:获得STR位点脂蛋白脂肪酶基因(LPL)在浙南汉族群体中的等位基因频率。方法:用Chelex法提取外周血DNA,PCR扩增短串联重复(STR)位点LPL后,用聚丙烯酰胺电泳系统进行分型。结果:在236个无关个体中共发现了四个等位基因,扩增片段为117-129 bp,等位基因频率最高为0.6758(117 bp),有十个核心重复序列,最低为0.0191(129 bp),有十三个核心重复序列。杂合率、个体识别率、非父排除率和多态性信息容量分别为0.4949、0.6996、0.1840、0.4497。家系分析表明按孟德尔规律遗传。结论:结果表明STR位点LPL是一个具有法医应用价值的位点,浙南汉族群体中的等位基因频率与武汉地区相比,差异无显著性。 Objective:To get the polymorphism of LPL in South Zhejiang Han population.Methods: Short tandem repeat (STR) locus LPL was analyzed by means of polymerase chain reaction, following polyacrylamide gel electrophoresis system.Results: Among 236 unrelated individuals from South Zhejiang Han population,4 alleles ranging from 117 - 129 bp were observed, The most common allele with a frequency 0.6758 was allele 117 bp with 10 core sequences, the rare allele was allele 129 bp with a frequency 0.0191 with 13 core sequences. The heterozygosity, the power of discrimination and the exclusion chance in paternity case and polymorphism information content were 0.4949,0.6996,0.1840 and 0.4497,respectively. Segregation studies revealed that LPL inherits in Medel' s Law. Conclusion: The result shows that locus LPL is highly informative and suitable for forensic application. Allele frequents are indifferent between South Zhejiang and Wuhan in Han population.
