双吲哚马来酰亚胺抑制鼻咽癌细胞端粒酶活性的实验研究 被引量:3

摘要 目的 探讨蛋白激酶C(PKC)抑制剂双吲哚马来酰亚胺对鼻咽癌细胞CNE2端粒酶活性的影响 ,为体内端粒酶活性的调节机制的研究提供理论基础。方法 用不同浓度双吲哚马来酰亚胺处理鼻咽癌细胞CNE2不同时间后 ,用台盼蓝染色法检测细胞生存能力 ,用PCR EIA法检测端粒酶活性。结果 PKC抑制剂双吲哚马来酰亚胺能显著地抑制鼻咽癌细胞CNE2端粒酶活性 ,且具有时间和剂量依赖性。该抑制剂不影响被处理细胞的生存能力 ,因为大多数被处理细胞能生存 (>80 % ) ,不同浓度双吲哚马来酰亚胺处理鼻咽癌细胞CNE2不同时间后细胞生存能力无统计学差异 (F =2 .4 4 ,P >0 .0 5 )。结论 双吲哚马来酰亚胺能显著抑制鼻咽癌细胞CNE2端粒酶活性 ,且具有时间和剂量依赖性。 Objective To investigate the effect of protein kinase C (PKC) inhibitor bisindolylmaleimideⅠon telomerase activity in nasopharyngeal carcinoma cell line CNE2, and to provide a theoretical basis for studying molecular mechanisms in the regulation of telomerase activity in vivo. Methods Nasopharyngeal carcinoma cell line CNE2 was treated with different concentration of Bisindolylmaleimide Ⅰ at different time intervals. Viability of the treated cells was determined by examination of cells stained with 0.2% Trypan blue. Polymerase chain reaction-based enzyme immunoassay (PCR-EIA) were used for assaying telomerase activity. Results The PKC inhibitor Bisindolylmaleimide Ⅰ was found to produce a significant inhibition of telomerase activity in the treated cells. The inhibition of telomerase activity by the PKC inhibitor appears to be specific since the treated cells were mostly viable (more than 80%). No significant difference was detected in the cell viability of CNE2 cells with treatment of different concentration of bisindolylmaleimide Ⅰ at different time intervals (F= 2.44, P> 0.05); and the PKC inhibitor produced dose-dependent and time-dependent inhibition of telomerase a ctivity of cell extracts. Conclusion These results indicate that PKC inhibitor bisindolylmaleimide Ⅰ is found to produce a significant inhibition of telomerase activity in the treated cells. The PKC inhibitor is able to produce dose-dependent and time-dependent inhibition of telomerase activity of cell extracts.
