近江牡蛎类立克次体在BALB/c小鼠中传代研究 被引量:3

摘要 从患病近江牡蛎的鳃组织中分离纯化得到类立克次体,然后在BALB/c小鼠体内感染传代。实验动物分组如下:(1)直接注射类立克次体;(2)注射用青霉素处理过的类立克次体;(3)同时注射青霉素处理的类立克次体和环磷酰胺;(4)注射灭菌的PBS。共传三代,分别于第3、7、11天解剖小鼠,采用实质性器官直接涂片、组织切片和透射电镜技术对小鼠体内的类立克次体进行检测。结果证明,近江牡蛎类立克次体对BABL/c小鼠是低毒或无毒的,具青霉素抗性;类立克次体虽可在其体内繁殖,但用环磷酰胺抑制免疫系统的BABL/c小鼠能更好地繁殖类立克次体。本实验成功建立了类立克次体在BABL/c小鼠体内连续感染传代繁殖的动物模型,找到了海洋贝类的RLO在陆地动物体内的培养方法,为RLO的分子免疫学及分子生物学的深入研究解决了获得RLO的动物模型问题,并且为研究RLO的致病机理提供了病理模型。这项工作在海洋贝类RLO研究中尚属首次。
